Total Internal Reflection Fluorescence (TIRF) Microscopy

The facility houses a dedicated Olympus cellTIRF microscope system (IX 81 inverted stand) equipped with a 100x/1.45 NA Plan Apochromat TIRF objective lens (in addition to 10x, 20x, 40x, and 60x lenses) is available. The system has DIC & phase optics, UV, FITC, TRITC, & quad filter sets, a low-profile stage incubator (to facilitate live cell imaging), Zero Drift Compensation autofocus, an Exfo X-cite illumination system and an BSI Prime back-illuminated sCMOS camera with image splitter form simultaneous two-channel imaging. The system has 405 nm, 489 nm diode, and 561 nm DPSS lasers, each with independent beam steering optics for three-channel TIRF and an AOTF to regulate beam intensity.

Two structured illumination microscopes (SIM) with TIRF capability. Furthermore, TIRF is also available on the Nikon N-SIM and Zeiss Elyra 7 structured illumination microscopes. TIRF may combined with or run independent of SIM. Both microscope are equipped with apochromat oil objectives (NA 1.49) providing 100x magnification, hardware autofocus system and 4 lasers (405, 488, 561 & 640 nm).
